Investigating the in vitro delivery and aerodynamic particle size distribution
of inhaled asthma medication using different valved holding chambers
VHC testing
We conduct comprehensive analysis of the aerodynamic particle size distribution (APSD) of salbutamol, inhaled corticosteroids, and other asthma medications when delivered through different Valved Holding Chambers (VHCs). Additionally, we study the influence of electrostatic charge on the VHCs.
State of the art methods
The aerodynamic particle size distribution (APSD) measurements are conducted using a Next Generation Cascade Impactor (NGI) in conjunction with a breath simulator. Our methos are in line with the international inhaler testing standards, making it suitable for assessing relevant flow rates. Most importantly, we use various pediatric and adult breathing patterns that closely emulate real-world scenarios offering results that can be readily applied in clinical practice.
Significance
The effective administration of inhaled medication has a far-reaching impact on a large number of patients, with the potential to enhance treatment efficacy, diagnostic precision, quality of life, and ultimately reduce healthcare expenses. The framework of our study aids in the identification of potential factors influencing in vivo medication delivery, informs the design of future in vitro studies, and contributes to the advancement of VHC design for the future.
Research highlights
Inhaled salbutamol's effective delivery is significantly influenced by the breathing patterns of small children and type of VHC used. Regulatory authorities should carefully evaluate and if necessary, restrict the approval and availability of VHCs that exhibit poor drug delivery characteristics. (Ojanperä et al.
